The ABI Prism 377 DNA Sequencing system is a significant upgrade of the familiar ABI 373A system. The flourescence detection component of the new system is the foundation for improved performance. The single photometer-filter wheel of the ABI 373A has been replaced by a CCD-based spectrometer. Serial scanning through each of four optical filters is functionally replaced by simultaneous monitoring of four virtual filter bins. The performance of the mechanical transport of the scanner across the platen of the sequencing gel is similar in each system, thus the ABI 377 should nominally realize a 4-fold acceleration of data acquisition. Improvements in the optical pathway of the new system increase the sensitivity of detection and quantification of the fluorescent labels.

The ABI PRISMĀ® 377 DNA Sequencer automatically analyzes DNA molecules labeled with multiple fluorescent dyes. After samples are loaded onto the system's vertical gel, they undergo electrophoresis, laser detection, and computer analysis. Electrophoretic separation can be viewed on-screen in real-time, and final data can be output in a variety of formats. The 377 system accepts gel plates in four different lengths for flexibility in sample analysis.
